SIRT7 is a deacetylase of N4-acetylcytidine on ribosomal RNA

چکیده
Abstract N-acetyltransferase 10 catalyzes RNA N4-acetylcytidine (ac4C) modifications and thus regulates stability translation efficiency. However, the deacetylase for ac4C is unknown. SIRT7 was initially identified as an NAD + -dependent protein plays essential roles in genome stability, circadian rhythms, metabolism, aging. In this study, we a of ribosomal (r)RNA first time found it to be -independent. Our data highlight important role rRNA modification suggest additional epitranscriptional regulation

The RNA acetyltransferase driven by ATP hydrolysis synthesizes N4-acetylcytidine of tRNA anticodon
The wobble base of Escherichia coli elongator tRNA(Met) is modified to N(4)-acetylcytidine (ac(4)C), which is thought to ensure the precise recognition of the AUG codon by preventing misreading of near-cognate AUA codon. By employing genome-wide screen of uncharacterized genes in Escherichia coli ('ribonucleome analysis'), we found the ypfI gene, which we named tmcA (tRNA(Met) cytidine acetyltr...
متن کاملThe role of the minor base N4-acetylcytidine in the function of the Escherichia coli noninitiator methionine transfer RNA.
The noninitiator methionine-accepting tRNA (tRNAmMct) of Escherichia coli contains N4-acetylcytidine at the wobble position of its anticodon. We have studied the role of this modified base in the function of tRNAmMct in protein synthesis. The biological activity of sodium bisulfite-treated tRNA,Met, which contains unmodified cytidine in place of A@-acetylcytidine but is otherwise identical to t...
متن کاملMethylation status of 13S ribosomal RNA from hamster mitochondria: the presence of a novel riboside, N4-methylcytidine.
The ribosomal RNA ("13S" RNA) of the small ribosomal subunit of hamster cell mitochondria has been found to have a distinctive pattern of methylated residues. Each molecule contained, on the average, approximately one residue of m4Cp, m5Cp and m5Up, and two residues of m62Ap. The natural occurrence of m4Cp has not previously been reported; we propose that this nucleotide is homologous to its ri...
متن کاملSIRT7-dependent deacetylation of CDK9 activates RNA polymerase II transcription
SIRT7 is an NAD+-dependent protein deacetylase that regulates cell growth and proliferation. Previous studies have shown that SIRT7 is required for RNA polymerase I (Pol I) transcription and pre-rRNA processing. Here, we took a proteomic approach to identify novel molecular targets and characterize the role of SIRT7 in non-nucleolar processes. We show that SIRT7 interacts with numerous proteins...
متن کاملLocation of ribosomal protein binding sites on 16S ribosomal RNA.
The distribution of ribosomal protein binding sites on the 16S ribosomal RNA molecule has been analyzed by limited ribonuclease hydrolysis of RNA-protein complexes, as well as by the interaction of individual proteins with RNA fragments purified from partial enzymatic digests. Of the six 30S subunit proteins known to interact directly with 16S RNA, proteins S4, S8, S15, S20, and, probably, S13 ...
